核膜的动态变化与细胞增殖调控/课题立项报告

摘 要:细胞核内外通过核膜进行着广泛的物质信息交流,这种交流对细胞基因的表达直至整个细胞生命活动均起着重要的调控作用。核膜的功能及细胞增殖调控研究是细胞生物学的前沿科学。核膜蛋白与核膜功能发挥及细胞增殖调控密切相关,其功能异常将导致人类重大疾病。该课题将围绕核膜动态变化及核膜功能中的关键科学问题,深入研究核膜融合及核孔复合体装配的分子基础和机理。主要研究内容包括参与核膜组装的关键蛋白,如,RanGTPase及其结合和调控蛋白、Importin-beta、Nucleoplasmin、HP1、LBR、Crm1、Pericentrin等在核膜动态变化中的作用,了解核膜膜泡的募集、融合以及核孔复合体装配的分子机制。

The Nuclear Envelope Dynamics and the Cell Proliferation Regulation /Project Report

Abstract:The material exchange between inside and outside of the nucleus,which is mainly through the nuclear envelope (NE), is important for the gene expressions and cell life. The NE proteins play crucial roles for the NE dynamics,the NE functions and the cell proliferation.Dysfunctions of the NE proteins are highly related to disorders of the NE dynamics and result in severe diseases.This project will mainly focuse on the the NE dynamics and functions, and the molecular mechanisms of the NE and nuclear pore complex assembly by investigating the functions of the key proteins in NE and nuclear pore complex assembly processes, including Ran GTPase and associated proteins, importin-beta, nucleoplasmin, HP1, LBR, Crm1 and pericentrin etc.
