高效液相色谱法测定断血流中香蜂草苷的含量

摘要 [目的]建立测定断血流中的香蜂草苷含量的方法，考察不同产地断血流中香蜂草苷的含量。[方法]采用Zorbax Eclipse XDB C-18柱（4.6 mm×150 mm，5.0 μm），柱温25 ℃，流动相为甲醇-水（48∶52），流速0.8 ml/min，检测波长285 nm。建立了高效液相色谱法测定断血流中香蜂草苷的含量。[结果]香蜂草苷在20.84～104.20 μg/ml的范围内线性关系良好（r=0.999 4），平均加样回收率（n=9）为98.63%（RSD=2.57%）。[结论]所建方法快捷简便、准确、重复性好，可作为断血流中香蜂草苷含量检测的方法。

HPLC Determination of Didymin in Duanxueliu Herb

Abstract [Objective] To establish a method by DADHPLC for determining the content of didymin in Duanxueliu herb from different habitats.[Method] Zorbax Eclipse XDB C18 chromatographic column（4.6 mm×150 mm，5.0 μm） and methanolwater（48∶52） as mobile phase were comployed，at a flow rate of 0.8 ml/min，a detecting wavelength at 285 nm.[Result] The standard curve of didymin was linear in the range of 20.84-104.2 μg/ml（r=0.999 4）.The average recovery of didymin was 98.63% with the RSD of 2.57% （n=9）.[Conclusion] This method is rapid，simple，sensitive，reliable，accurate and suitable for the analysis and determination of didymin in Duanxueliu herb.

斷血流为唇形科植物荫风轮Clinopodium polycephalum （Vaniot）C.Y.Wu et Hsuan 或风轮菜Clinopodium chinensis（Benth.）O.Kuntze 的干燥地上部分，具有收敛止血之功效，主要用于治疗崩漏、尿血、鼻衄、牙龈出血、创伤出血等出血症及单纯性紫癜、原发性血小板减少性紫癜等症^[1]。中国药典2010版一部中将醉鱼草皂苷IVb作为其主要活性成分进行鉴别检测^[2]。除醉鱼草皂苷外，断血流中还含有多种活性成分，主要是黄酮及其苷、三萜及其苷类以及环烯醚萜苷类等。近年来，其中的黄酮苷——香蜂草苷开始引起大家的研究热情。香蜂草苷最早由Wagner等于1967年在同科美国薄荷属（Monarda）植物美国薄荷（Monarda didyma）的叶中发现^[3]，戴金瑞在1983年首次从风轮菜中分离并鉴定该化合物^[4]，陈靖宇等1997年首次从荫风轮中分离并鉴定该化合物（图1）^[5]。最新的药理研究发现香蜂草苷可通过抑制癌基因 N-Myc 以及上调Raf激酶抑制蛋白（RKIP）诱导神经母细胞瘤细胞凋亡^[6]，使其可能成为一个非常有潜力的治疗神经细胞瘤的先导化合物。同时，该化合物也显示出了对非小细胞肺癌有一定的治疗效果^[7]。……
