摘要:研究了暴露在不同浓度高效氯氰菊酯下草鱼(Ctenopharyngodon idellas)鳃、肝胰脏和肾脏组织中碱性磷酸酶(Alkaline phosphatase,AKP)的活性变化。试验中高效氯氰菊酯浓度设5组(0、0.5、1.0、3.0和5.0 μg/L),每组分别于1、5和12 d时取样,测定鳃、肝胰脏和肾脏组织中AKP的活性。结果表明,0.5 μg/L高效氯氰菊酯暴露1 d时,鳃中AKP活性显著下降(P<0.05),肝胰脏和肾脏中的AKP活性无显著变化(P>0.05);暴露5 d时,鳃和肾脏中AKP活性显著或极显著降低(P<0.05,P<0.01),肝胰脏中AKP活性无显著变化(P>0.05);暴露12 d时,鳃和肾脏中AKP活性极显著下降(P<0.01),肝胰脏中AKP活性无显著变化(P>0.05)。1.0 μg/L高效氯氰菊酯暴露1 d,肝胰脏和肾脏中AKP活性无显著变化(P>0.05),鳃中AKP活性显著或极显著下降(P<0.05,P<0.01);暴露5 d时,肾脏中AKP活性显著或极显著下降(P<0.05,P<0.01),肝胰脏中AKP活性无显著变化(P>0.05);暴露12 d,鳃、肝胰脏和肾脏中AKP活性显著下降(P<0.01)。3.0 μg/L和5.0 μg/L高效氯氰菊酯显露1、5和12 d,所测定组织中AKP活性均显著或极显著下降(P<0.05,P<0.01)。

Changes of Alkaline Phosphatase Activity in Organs of Grass Carp Exposed to the Different Beta-cypermethrin Concentration

Abstract: To study dynamic changes of alkaline phosphatase activity(AKP) in gill, hepatopancreasand kidney of grass carp(Ctenopharyngodon idellas)exposed to the different Beta-cypermethrin concentration can be helpful to understand of Beta-cypermethrin effect on AKP. In this study,grass carps were randomly divided into five groups including control group and four different Beta-cypermethrin concentration groups (0.5、1.0、3.0 and 5.0 μg/L).Each was in replication. Biochemistry methods were used to measure AKP activity of gill,hepatopancreas and kidney sampled on day 1,5 and 12,respectively. The results were as follows: contrast to the control,for 0.5 μg/L,AKP activity of gills was sharply reduced(P<0.05), but that of hepatopancreas and kidneys didnt significantly change (P>0.05)on day 1,then AKP activity of gills and kidneys sharply reduced(P<0.01,P<0.05),but no significant change happened for that of hepatopancreas(P>0.05) on day 5. Finally that of gills and kidneys significantly decreased (P<0.01),and that of hepatopancreas returned to the normal level on day 12.For 1.0 μg/L,AKP activity of gills was sharply reduced(P<0.01,P<0.05),but that of hepatopancreas and kidneys didnt significantly change (P>0.05)on day 1,then AKP activity of serum significantly increased(P<0.01),that of spleens and kidneys were sharply reduced(P<0.01,P<0.05),but that of hepatopancreas didnt significantly change(P>0.05) on day 5. Finally AKP activity of serum didnt sharply change (P>0.05),that of gills,hepatopancreas,spleens and kidneys significantly decreased (P<0.01)on day 12. For 3.0 μg/L and 5.0 μg/L,AKP activity of serum,gills,hepatopancreas,spleens and kidneys significantly decreased(P<0.01,P<0.05).

高效氯氰菊酯是一种高效低毒的拟除虫菊酯类农药,因其对哺乳动物、鸟类的低毒性和对有害昆虫的高效杀灭性而被广泛的应用于农作物、园艺植物、家庭卫生害虫的防治[1]。在水产养殖方面也被作为有机磷农药的替代品用来防治寄生虫和控制有害水生昆虫的过度增殖[2,3]。由于过量使用,雨水冲刷使高效氯氰菊酯通过地表径流进入河道、湖泊等水体,导致非杀灭目标的水生无脊椎动物、鱼类等对神经毒素类农药非常敏感的生物受到影响[4,5]。研究表明,拟除虫菊酯对鱼类的毒性是哺乳动物和鸟类的1 000倍[6]。目前,关于高效氯氰菊酯对鱼类的影响方面的研究报道很多,主要集中于对鱼类的毒性研究方面[7,8],对鱼类的代谢酶活性的影响方面也有一定量的报道[9,10]。碱性磷酸酶(Alkaline phosphatase,AKP)在生物体内核酸、蛋白质和脂类等物质代谢中起着重要的作用,但鲜见高效氯氰菊酯对鱼类AKP活性影响方面的报道。高效……
