摘要:凝集素类受体蛋白激酶(Lectin-like receptor kinase,LecRK)是一类植物特有并分布广泛的蛋白激酶。采用RT-PCR 从水稻(Oryza sativa L.)品种B5中克隆了OsLecRK基因靠近翻译起始位点下游的530 bp 的特异基因片段,将该片段以正、反向分别连入pKANNIBAL载体,从而获得pKAN-OsLecRK-RNAi的中间表达载体,进而将其克隆到植物双元表达载体pCAMBIA1301中,构建dsRNAi表达载体pCAMBIA-OsLecRK-dsRNAi。经酶切和测序鉴定正确后,利用农杆菌介导转化水稻,通过抗性筛选和标记基因hyg进行PCR鉴定,筛选出12株转基因水稻植株。

Construction and Genetic Transformation of RNA Interference Vectors
for OsLecRK Gene of Oryza sativa L.

Abstract: Lectin-like receptor kinase(lecRK) is a family of proteins which is unique and universal in plants. A 530 bp gene specific fragment of OsLecRK was amplified using RT-PCR method from rice variety B5 and this fragment was linked both in sense and antisense directions to the vector of pKANNIBAL as junction fragment. The resulting construct was then cloned into plant binary expression vector pCAMBIA1301 to form dsRNAi expression vector pCAMBIA-OsLecRK-RNAi. After confirmation by enzyme digestion and sequencing analysis, this pCAMBIA-OsLecRK-RNAi was transferred into rice by agrobacterium-mediated transformation method. A total of 12 regenerated transgenic lines were screened through PCR identification of hyg gene.

水稻(Oryza sativa L.)是世界的主要粮食作物之一,全球超过一半人口以水稻作为主食[1]。水稻是中国最重要的口粮作物,常年种植面积约3 000万hm2,占世界水稻种植总面积的1/4。因此,水稻的种植安全与中国农村经济发展和口粮安全密不可分。
凝集素类受体蛋白激酶(lectin-like receptor kinase,LecRK)是一类植物特有并分布广泛的蛋白激酶。自1996年从拟南芥中第一次分离得到该家族成员Ath.lecRK1后,人们已从多种植物中克隆到多个该类蛋白基因。研究表明,LecRK基因对植物生长发育有一定的作用。拟南芥sgc是一种LecRK基因失活的突变体,由T-DNA插入At3g53810而引起,突变体sgc由花粉时期开始,所有的花粉粒都开始变形和萎缩,从而引起雄性不育[2]。LecRK基因还广泛参与植物与生物和非生物胁迫的互作。LecRK胞外的凝集素受体结构域一直被认为是与根瘤菌的共生和病原体的抵抗相关。最近的研究也证明了这个观点,新发现的水稻抗稻瘟病Pi-d2就是一种包含有甘露糖特异的凝集素受体蛋白激酶[3]。……
