鼻咽癌组织中miR-200a的表达及意义

摘要:目的观察鼻咽癌和鼻咽黏膜炎性组织中microRNA-200a(miR-200a)的表达变化,分析其与鼻咽癌发生发展的关系。方法取鼻咽癌组织39例(观察组)、鼻咽黏膜炎性组织12例(对照组)。采用实时荧光定量PCR方法检测两组miR-200a表达,比较不同临床病理参数(包括年龄、性别、T分期、N分期、临床分期)的鼻咽癌患者miR-200a表达差异。结果观察组和对照组miR-200a的相对表达量分别为0.539±0.281、0.744±0.358,观察组较对照组明显下降(P<0.05)。鼻咽癌组织中miR-200a表达: T3、T4期低于T1、T2期,N2、N3期低于N0、N1期,临床分期Ⅲ、Ⅳ期低于Ⅰ、Ⅱ期(P<0.05或<0.01)。结论miR-200a在鼻咽癌组织中的表达明显下调,其参与了鼻咽癌的发生发展过程。

Expression and clinical significance ofmiR-200a in nasopharyngeal carcinoma

Abstract:Objective To observe the expression changes in levels ofmiR-200a in the tissues of nasopharyngeal carcinoma(NPC)and nasopharyngitis and to analyze its relationship with the occurrence anddevelopment of NPC.Methods We took 39 cases of nasopharyngeal carcinoma tissues as the observation group and 12 cases of nasopharyngitis tissues as the control group.The real-time fluorescent quantification PCR was used todetect the expression levels ofmiR-200a in the two groups.Thedifferential expression levels ofmiR-200a in NPC patients with clinicopathological parameter(including age,gender,T stage,N stage and clinical grade)were compared.Results The relative expression ofmiR-200a in the observation group and the control group was 0.539±0.281 and 0.744±0.358,respectively.The expression ofmiR-200a in the observation group was significantlydecreased as compared with that of the control group(P<0.05).The expression ofmiR-200a in the observation groupdecreased with T stage,N stage and the increasing clinical stages(P<0.05 or P<0.01).Conclusion The expression ofmiR-200a in the NPC tissues isdown-regulated whichmay participate in the pathogenesis anddevelopment of NPC.

鼻咽癌(NPC)是我国南方地区常见的恶性肿瘤之一,其发病率在头颈部恶性肿瘤中居首位[1]。研究发现,microRNA-200a(miR-200a)在多种肿瘤中表达下调,在肿瘤发生、发展过程中具有抑癌基因的功能[2]。有关miR-200a在鼻咽癌中表达的情况少见报道。本研究采用Real-time PCR方法检测鼻咽癌组织中miR-200a表达,探讨其在鼻咽癌发生发展中的作用。
1.1临床资料选取2014年4~6月在广西医科大学第一附属医院行活组织病理检查的39例鼻咽癌患者(观察组)和12例鼻咽黏膜炎性患者(对照组),均经病理检查确诊。观察组男24例、女15例,年龄25~74岁、中位年龄45岁。患者均为初发,未接受化疗、放疗及其他针对肿瘤的相关治疗。……
