miRNA在中医药防治结直肠癌中的研究进展

摘 要 在结直肠癌的发病机制中，miRNA表达失调与中医“正虚邪实”的病机异途同归，miRNA的表达及调控可能是中医药发挥抗肿瘤疗效的作用机制之一。全文将miRNA调控机制与中医“扶正祛邪”理论相结合，探讨miRNA调控网络与中医理论之间的联系，并从中药单体及活性成分和中药复方两个方面阐述以miRNA为靶点的中医药防治结直肠癌的研究现状。

Research progress of traditional Chinese medicine in the prevention and treatment of colorectal cancer with miRNA

ABSTRACT In the pathogenesis of colorectal cancer， disorder of miRNA expression coincides with “ healthy energy deficiency and evil excess “ in the pathogenesis of traditional Chinese medicine （TCM）. The expression and regulation of miRNA may be one of the mechanisms of TCM in against the cancer. In this paper， the relationship between the regulation network of miRNA and TCM is discussed based on the combination of the regulation mechanism of miRNA and the TCM theory of“ strengthen body resistance and eliminating evil ”. And the research status of TCM in the prevention and treatment of colorectal cancer with miRNA as the target is introduced from two aspects of monomer and active ingredient extracted from Chinese medicine and Chinese medicine compound.

结直肠癌新发病例在全世界男性、女性中分别排名第3位及第2位，死亡率分别排名第4位及第3位[1]。我国东部地区结直肠癌发病率及死亡率最高，可能与东部发达地区人群生活方式明显西化有关；西部地区发病率、死亡率与东部地区相似，而中部地区的发病率、死亡率则均较低。大规模人群筛查是降低结直肠癌发病率与死亡率的重要途径，目前的筛查方法主要包括粪隐血试验、血清癌胚抗原检测、结肠镜检查、气钡双重造影检査等。结直肠癌早期诊断率低、复发转移率高，且缺乏有效治疗手段，是导致患者高病死率的主要原因[2]。寻找结直肠癌发病机制中的关键调控分子，是提高其临床诊治水平的关键。microRNA （miRNA）是存在于真核细胞中具有进化保守性的一族非编码小片段RNA，大小为21～23 nt，通过与其序列互补的靶基因mRNA的碱基互补配对影响mRNA的稳定性或抑制其翻译，实现对蛋白表达的调控。miRNA控制着细胞的分化、增殖和程序性细胞死亡等多种重要生理过程，并与多种疾病如肿瘤相关[3]。……
