地塞米松诱导小鼠高血压的副作用研究

【摘要】 目的 探討地塞米松对小鼠血压和心率的影响,寻找制备动物高血压模型的药物。

方法 取KM小鼠40只随机分为正常对照组(给予0.9% NaCl注射液)和地塞米松低、中、高浓度组(分别给予1.25 mg/kg、2.5 mg/kg、5.0 mg/kg地塞米松注射液),肌肉注射每天一次(用量按小鼠体重10 mL/kg计算),连续14天。各组小鼠在末次用药后20 min测定收缩压、舒张压和心率。

结果 正常对照组和地塞米松低浓度组的实验后收缩压、舒张压、心率与实验前比较差异均无统计学意义(P>0.05)。地塞米松中浓度组和高浓度组实验后的收缩压、舒张压均明显高于实验前和正常对照组(P<0.05或0.01),地塞米松高浓度组实验后的收缩压、舒张压均明显高于地塞米松低浓度组、中浓度组(P<0.01)。正常对照组和地塞米松低浓度组、中浓度组的实验前后心率两两比较差异无统计学意义(P>0.05)。地塞米松高浓度组的实验后心率与正常对照组和地塞米松低浓度组、中浓度组比较差异无统计学意义(P>0.05),但明显高于其实验前(P<0.05)。

结论 地塞米松可加强心血管兴奋引起升高血压的作用,此作用存在量效关系;地塞米松用量达一定高浓度可加快心率。

【Abstract】 Objective To explore the effect of dexamethasone on blood pressure and heart rate in mice,and to search for drugs for the preparation of animal hypertension model.

Methods 40 KM mice were randomly divided into normal control group(given 0.9% NaCl injection) and dexamethasone low concentration group,dexamethasone medium concentration group and dexamethasone high concentration group(given 1.25 mg/kg,2.5 mg/kg and 5.0 mg/kg dexamethasone injection,respectively).Intramuscular injection was performed once a day(dosage was calculated according to 10 mL/kg of mouse body weight) for 14 consecutive days.And then,systolic blood pressure,diastolic blood pressure and heart rate were measured 20 min after the last administration in each group.

Results There was no statistically significant difference in systolic blood pressure(SBP),diastolic blood pressure(DBP) and heart rate(HR) between the normal control group and dexamethasone low concentration group before and after experiment(P>0.05).After experiment,the systolic and diastolic blood pressure of dexamethasone in the middle and high concentration groups were all significantly higher than those before experiment and those in the normal control group(P<0.05 or 0.01),and those in the dexamethasone high concentration group were significantly higher than those in the dexamethasone low concentration group and dexamethasone medium concentration group(P<0.01).There was no statistically significant difference in heart rates between the normal control group,the dexamethasone low concentration group and the dexamethasone medium concentration group before and after experiment(P>0.05).After experiment,there was no statistically significant difference in heart rate between the dexamethasone high concentration group, the normal control group,the dexamethasone low concentration group and the middle concentration group(P>0.05),but they were all higher than those before experiment(P<0.05).

本研究结果警示,使用地塞米松治疗的患者应加强对血压的监测,高血压、冠心病、心脑血管意外的患者慎用。
