Wnt信号通路与卵巢癌治疗新进展

【摘要】Wnt通路是一条高度保守的传导通路,Wnt信号通路异常激活与肿瘤发生发展有密切关系。近年来,不少研究证明卵巢癌中存在异常激活的Wnt通路信号蛋白。该文复习了卵巢癌与Wnt通路相关的新近研究进展,对Wnt信号通路、其在卵巢癌发生发展的作用及Wnt通路中潜在的卵巢癌治疗新靶点做一综述,旨在为卵巢癌的临床治疗提供新思路。

【Abstract】Wnt signaling pathway is a highly conserved pathway. Abnormal activation of the Wnt signaling pathway is intimately correlated with the incidence and progression of tumors. In recent years, accumulated studies have proven that the proteins related to the abnormally-activated Wnt signaling pathway are expressed in ovarian cancer. In this article, the latest research progress on the ovarian cancer and the Wnt signaling pathway was reviewed, the role of the Wnt signaling pathway in the incidence and progression of ovarian cancer and the potential novel therapeutic targets for ovarian cancer in the in Wnt signaling pathway were summarized, aiming to provide novel ideas for the clinical treatment of ovarian cancer.

卵巢癌是女性生殖系統恶性肿瘤之一,其病死率居于妇科恶性肿瘤首位。卵巢癌中80%的病理类型为上皮性卵巢癌(EOC)。输卵管分泌细胞是大多数卵巢癌的起源细胞。卵巢癌早期症状隐匿,发现时多为中晚期,预后较差。Wnt是一类分泌型糖蛋白,通过自分泌或旁分泌发挥作用。Wnt信号通路具有调节干细胞生长和器官发育的作用,其异常激活会诱导肿瘤的发生、发展和转移。本文通过总结Wnt通路的新近临床研究进展,分析其在卵巢癌靶向治疗中的作用,旨在为卵巢癌的临床治疗提供新思路。

一、Wnt信号通路

1982年Nusse等首先发现了Wnt家族,之后人们对Wnt信号通路的认识不断加深,目前Wnt信号通路已基本被阐明,其中包括经典的Wnt/β-连环蛋白(catenin)信号通路,非经典的Wnt通路,后者又包括Wnt-Ca2+信号通路、平面细胞极性(PCP)信号通路。Wnt信号通路相关基因突变与胃肠癌、白血病、黑色素瘤、乳腺癌及卵巢癌等发生有密切关系[1]。
