盐酸丙美卡因滴眼液有关物质检测和含量检测方法研究

摘要：目的：研究确认盐酸丙美卡因滴眼液有关物质检测和含量检测方法的可行性。方法：采用高效液相色谱法，对盐酸丙美卡因滴眼液有关物质和含量的检查方法进行研究。结果：通过系统适用性试验、回收率试验、精密度试验，确定了盐酸丙美卡因滴眼液有关物质和含量各项检测条件的合理性。结论：可采用高效液相色谱法进行本品盐酸丙美卡因的有关物质和含量检测。

[Abstract] Objective： To confirm the feasibility of the detection of related substance and its content in proparacaine hydrochloride eye drops.Methods： To study the detection of related substance and its content in proparacaine hydrochloride eye drops through high performance liquid chromatography （HPLC）. Results： It is determined that the detection method for related substance and its content in proparacaine hydrochloride eye drops is appropriate by the system suitability test， recovery and precision test. Conclusion： HPLC can be used to detect the related substance and its content in proparacaine hydrochloride eye drops.

盐酸丙美卡因滴眼液有关物质和含量检测方法的可行性，将直接影响到产品上市后的有效性、安全性和质量稳定性。故本次采用高效液相色谱法，通过系统适用性试验、回收率试验、精密度试验，确定了盐酸丙美卡因滴眼液有关物质和含量各项检测条件的合理性。从而确认盐酸丙美卡因的有关物质和含量检测检测方法。

1 仪器与试药

1.1 仪器

岛津LC-10ATVP泵，岛津SPD-10AVP检测器，TL-9900色谱数据工作站，MS205DU电子天平

1.2 试药

磷酸二氢钾（分析纯，北京益利精细化学品公司），庚烷磺酸钠（色谱纯 进口），三乙胺（色谱纯 进口），甲醇（色谱纯，北京益利精细化学品公司）

2.有关物质检测方法研究

采用高效液相色谱法，对本品有关物质的检查方法进行研究，照中国药典2015年版四部0512[1]。

2.1色谱条件

色谱柱：Inertsil Ph-3苯基柱，250mm×4.6mm，5m

参照英国药典盐酸丙美卡因质量标准 [2]，有关物质检查色谱条件以磷酸盐缓冲液（0.05mol/L磷酸二氢钾、0.02mol/L庚烷磺酸钠，三乙胺调pH值至5.2）-甲醇（38：62） 为流动相。……
