威灵仙-白花蛇舌草药对抗癌作用机制研究

摘  要：探讨威灵仙-白花蛇舌草药对治疗癌症的作用机制.从HERB与Swiss ADME两个数据库中筛选出威灵仙-白花蛇舌草药对中活性成分、作用靶点、癌症疾病靶点、活性成分与疾病的共同靶点，筛选出重要的活性成分与关键靶点，将活性成分与关键靶点进行分子对接验证.关键靶点为ESR1，重要活性成分为槲皮素、阿魏酸和恩贝林，这三种药物的活性成分均与疾病靶点蛋白有着较好的结合性.威灵仙-白花蛇舌草药对可能对治疗癌症疾病起着重要作用.

Abstract：Exploring the Mechanism of couplet medicines of Clematis chinensis Osbeck and Hedyotis diffusa Treatment for Cancer.The HERB and Swiss ADME databases were used to screen active ingredients， target proteins， and common targets of Clematis chinensis Osbeck and Hedyotis diffusa for cancer treatment， and important active ingredients and key targets were identified and verified through molecular docking.The key target is ESR1， and the important active ingredients are Quercetin， Ferulic acid， and Embelin.All three active ingredients have good binding properties with the disease target protein.Couplet medicines of Clematis chinensis Osbeck and Hedyotis diffusa may play an important role in the treatment of cancer.

癌癥是由于生物体某个基因发生突变导致的[1-2]，中草药如威灵仙（Clematis chinensis Osbeck），常与其他中药配伍治疗疾病；白花蛇舌草（Hedyotis diffusa）是南方较为常见的中草药，有着消炎、解毒、抗癌肿等多种药理作用，具有较好的使用价值.[1-4]本文以威灵仙-白花蛇舌草为药对，采用活性成分筛选法[5]，通过网络药理学与分子对接实验，探讨威灵仙-白花蛇舌草药对的抗癌机制.

1 材料与方法

威灵仙-白花蛇舌草对活性成分的收集和筛选 在数据库本草组鉴（http：//drug.ac.cn/，HERB）中分别搜索威灵仙、白花蛇舌草的成分，选择OB score ≥ 30为筛选条件，分选出主要成分.在 Swiss ADME（http：//www.swissadme.ch/）中，以药代动力学里的胃肠道吸收（GI absorption）为“High”的结果，及类药性（Druglikeness）五个预测（Lipinski，Ghose，Veber，Egan，Muegge）的结果中有2个及以上是“Yes”为筛选条件，对HERB中收集的活性成分进行筛选，筛选得到最终的活性成分.
