摘要:通过ISSR分子标记技术,对采自河北石家庄、保定、邢台、邯郸、张家口的11个杠柳(Periploca sepium Bunge)样品进行遗传多样性分析。结果表明,从100条通用引物中共筛选出34条适用于杠柳ISSR研究的多态性引物;对11个供试样品进行PCR检测,进一步筛选出8条多态性好、稳定性高的引物,共检测出79个ISSR位点,多态性位点63个,占79.7%;遗传相似系数(GS)变化范围在0.454 5~0.854 5,8对多态性引物多态信息含量值变化范围为0.747~0.884,平均为0.827;11份供试样品聚类分为三大类,聚类相对复杂,呈一定的地域相关性;ISSR分子标记适用于杠柳遗传多样性研究,且杠柳遗传多样性丰富。

Genetic Diversity Analysis of Periploca sepium Bunge in Hebei Province by ISSR Markers

Abstract:ISSR makers was used to analyze the genetic diversity of 11 Periploca sepium Bunge samples from Shijiazhuang, Handan, Xingtai, Zhangjiakou, Baoding. The results showed that 34 polymorphic primers were selected from 100 general primers for ISSR analysis. 11 samples were tested for PCR, and 8 primers with good polymorphism and stability were screened out, 79 ISSR loci were amplified by 8 primers, among which 63 loci were polymorphic, accounting for 79.7%. Genetic similarity coefficient varied in the range of 0.454 5~0.854 5. the change range of PIC values of polymorphic primers was 0.747~0.884, the average was 0.827. 11 samples were clustered into 3 groups, and the cluster was relatively complex, the accessions were significantly related to the geographical. ISSR molecular markers were applied to the study of genetic diversity of Periploca sepium Bunge, and the genetic diversity was rich.

杠柳(Periploca sepium Bunge)又叫北五加皮、桃枝子、羊奶子、羊角桃等,是蘿藦科杠柳属植物。在河北全省均有分布,主要集中于太行山山区,道路两旁以及田间荒坡,其根皮为传统中药材,有祛风湿、利水消肿、强筋骨的作用[1],其特性为喜光、耐寒、耐旱、耐荫、耐盐碱。根系分布深,呈丛生状态[2]。杠柳具有广泛的适应性,是优良的固沙、水土保持树种,不仅是优良的高速公路边坡绿化树种,同时也是煤矸石山生态恢复的可自然定居树种[3]。近年来,随着杠柳药理作用不断被开发,市场需求增加,出现过度采挖,而其种苗繁育迟缓,不利于进一步开展资源保护利用以及人工培植规划[4]。目前,国内外对杠柳研究多集中于生理生态及药理生态的研究,DNA分子水平研究鲜见报道。本……
