摘要 目的:观察松郁安神方对对氯苯丙氨酸(Para-chlorophenylalanine,PCPA)致失眠模型大鼠血浆中内源性睡眠促进物质前列腺素D2(Prostaglandin D2,PGD2)的影响。方法:采用腹腔注射对氯苯丙氨酸(PCPA)建立失眠大鼠模型,用高、中、低剂量松郁安神方进行治疗,通过酶联免疫吸附法(ELISA)测定大鼠血浆中前列腺素D2(PGD2)的含量。结果:中、高剂量松郁安神方可提高PCPA失眠模型大鼠血浆中PGD2含量(P<0.05或P<0.01)。结论:松郁安神方改善睡眠的机制之一可能是通过调节内源性睡眠促进物质PGD2发挥作用。

Effect of Songyu Anshen Formula on Levels of Plasma Prostaglandin D2 (PGD2 ) in Insomnia Rats Induced by Para-chlorophenylalanine(PCPA)

Abstract Objective:To observe the effect of Songyu Anshen Formula on the endogenous sleep-promoting substance PGD2 in the plasma of insomnia rats induced by PCPA.Methods:The rat model of insomnia was established by intraperitoneal injection of PCPA. The rats were given Songyu Anshen formula intragastrically, and the content of PGD2 in plasma was determined by enzyme-linked immunosorbent assay (ELISA). Results:The content of PGD2 in the plasma of PCPA could be increased by the middle and high dose of Songyu Anshen formula (P<0.05 or P<0.01 ) . Conclusion:One of the mechanisms of Songyu Anshen in improving sleep quality may be through the regulation of the endogenous sleep-promoting substance PGD2.

睡眠和觉醒的发生及转换可能是通过脑内神经递质和内源性睡眠促进物质共同作用调节的。其中前列腺素D2(PGD2)和腺苷是最强效的内源性睡眠促进物质,参与生理性睡眠觉醒调节[1]。松郁安神方根据疏肝安神法,以甘松、郁金、玫瑰花为君药,配伍酸枣仁、首乌藤、丹参、生龙骨、珍珠母、合欢皮组方,具有重镇安神之功效,在临床上治疗失眠疗效显著[2-3]。本试验通过观察松郁安神方对内源性睡眠促进物质PGD2的影响,探讨松郁安神方改善睡眠的作用机制。
1 材料
1.1 试验动物 清洁级SD大鼠36只,雄性,体质量(230±10)g,购自上海斯莱克试验动物有限责任公司(生产许可证号:SCXK(沪)2017-0005;合格证编号:20170005004461)。动物置于安静、恒温、恒湿、12 h/12 h明暗光照环境中饲养,每天给予充足食物和水。试验前采用随机数字法将大鼠分为6组,分别为空白对照组、PCPA失眠组、中药高、中、低剂量组、酸枣仁汤阳性对照组,每组6只。
1.2 主要试剂与仪器 Rat PGD2 ELISA Kit(上海酶联生物科技有限公司,批号:Dec2018);对氯苯丙氨酸(PCPA)(Sigma公司,批号:SHBH7873);中药饮片(福建鹭燕药业股份有限公司);酶标仪(美国BioTek,800TS)。……
